Affinity purification of DNA and RNA from environmental samples with peptide nucleic acid clamps

Article Abstract:

The development of peptide nucleic acid clamps for extracting nucleic acids from soils with low biomass is described. The polymerase chain reaction has revitalized environmental microbiology by allowing researchers to analyze nucleic acid samples from soil rather than culturing organisms.

Application of the 5' fluorogenic exonuclease assay (TaqMan) for quantitative ribosomal DNA and rRNA analysis in sediments

Article Abstract:

Environmental samples contain many substances that may interfere with fluorescence detection using the TaqMan assay, including organic contaminants, metals, chelators, and humic acids. Researchers offer tips for preventing this interference when using TaqMan to analysis DNA and RNA in sediments and other environmental samples.

Direct detection of 16S rRNA in soil extracts by using oligonucleotide microarrays

Article Abstract:

Researchers show that microbial rRNA can be detected in soil samples using oligonucleotide microarrays without using PCR. Using a chaperone-detector probe strategy, they were able to detect 16S rRNA from Geobacter chapellei directly in a soil extract with no further purification or removal of other soil constituents.
