Extragenic suppressors of a dynein mutation that blocks nuclear migration in Aspergillus nidulans

Article Abstract:

The choice and characterization of extragenic nudA1 mutation suppressors helps detect other proteins exhibiting indirect or direct interactions with the cytoplasmic dynein heavy chain in Aspergillus nidulans. Five extragenic nudA1 suppressors (snaA-E) are identified through genetic analysis of 16 revertants exhibiting cold or dimethyl sulfoxide sensitivity. Nuclear migration phenotype of the sna mutants is studied by germinating, fixing and staining of nuclei of the conidia of each complementation group. The nudA1 nuclear migration defect is incompletely inhibited in the sna mutants.

An alpha tubulin mutation suppresses nuclear migration mutations in Aspergillus nidulans

Article Abstract:

The genes that mediate nuclear migration in Aspergillus nidulans were identified by isolating the extragenic suppressors of the nudF gene through UV mutagenesis. Molecular techniques identified one extragenic suppressor that is caused by a deletion in tubA alpha tubulin gene. The mutation destabilized microtubules, altered microtubule staining and conferred cold-sensitivity. Treatment with low concentrations of benomyl suppressed heat-sensitive mutations in nudA, nudC, nudF and nudG mutations.

Identification of developmental regulatory genes in Aspergillus nidulans by overexpression

Article Abstract:

Growth inhibition and development in Aspergillus nidulans is caused by the overexpression of numerous developmental regulatory genes. These developmental regulators are identified by the construction of a nutritionally inducible Aspergillus nidulans expression gene library. The genes consist of small, random, genomic DNA fragments. Sixty six different developmental genes have been identified by this method.
