Hydrodynamic effects on microcapillary motility and chemotaxis assays of Methylosinus trichosporium OB3b

Article Abstract:

The Palleroni microcapillary assay was evaluated with regard to the chemotactic behavior of Methylosinus trichosporium OB3b, an obligate methanotroph which degrades a number of chlorinated hydrocarbons. The results showed that the Palleroni microcapillary assay is subject to abiotic hydrodynamic effects arising from a density-induced convection. The source of this convection was most probably the fluid density difference between thee cell suspension and the microcapillary buffer. This procedure is more useful when evaluating the behavior of highly motile strains of bacteria.

Transformation kinetics of chlorinated ethenes by Methylosinus trichosporium OB3b and detection of unstable epoxides by on-line gas chromatography

Article Abstract:

On-line gas chromatography reveals that chlorinated ethene oxidation by Methylosinus trichosporium OB3b expressing methane monooxygenase or soluble methane monooxygenase leads to corresponding epoxide generation. Degradation of trichloroethane, trans-1,2-dichloroethane, vinyl chloride and cis-1,2- dichloroethane forms corresponding epoxides. There is a degradation of the trichloroethane and vinyl chloride epoxides according to first-order kinetics. Acetylene and methane suppresses the degradation of the cis-1,2- dichloroethane epoxide.

Degradation of trichloroethylene by methanol-grown cultures of Methylosinus trichosporium OB3b PP358

Article Abstract:

A study was conducted on the chemostat growth of PP358, a mutant strain of Methylosinus trichosporium OB3b. The strain was grown in modified nitrate minimal salt mediums containing 0.1-0.5% methanol. PP358 exhibited excellent trichloroethylene degradation ability in all growth conditions. PP358 grown in methanol-limiting conditions displayed lower degradation rates compared with that under oxygen-limiting conditions.
