Kinetics of photoacclimation in cultures of Chromatium vinosum DSM 185 during shifts in light irradiance

Article Abstract:

The kinetics of photoacclimation of continuous cultures of Chromatium vinosum DSM 185 were examined during transitions in light irradiance until steady state was achieved. Bacteriochlorophyll synthesis stopped for four hours when irradiance was shifted from four to 67 microE m(super -2) s(super -1). During this stage, pigments were progressively diluted in the newly created biomass, leading to a reduced specific pigment content. The shift from high to low irradiance was accompanied by an immediate decline in the specific growth rate to virtually zero as well as comparatively dramatic reductions in the specific rates of sulfur and sulfide oxidation and in the specific rate of glycogen accumulation.

Measurement of light absorption and determination of the specific rate of light uptake in cultures of phototrophic microorganisms

Article Abstract:

The specific rate of light uptake by microorganisms is calculated by dividing the rate of light absorbed, by the biomass in the culture. The rate of light absorbed is determined by measuring the rate of light output from a pure culture and a culture containing cells. The difference in light output is due to absorption by the microorganisms. The method fails to consider the light output from the top and bottom of the container which causes a small error in dense culture but a significant error in dilute cultures. The light absorbed is assumed to be within the range of the sensitivity of the light meter.

Description o a redox-controlled sulfidostat for the growth of sulfide-oxidizing phototrophs

Article Abstract:

Simulation of the growth of phototrophic sulfur oxides under constant concentrations of hydrogen sulfide in the culture and under conditions of changing illumination was conducted to represent the redox of the culture. Variations in biological activity were associated with variations in redox potential, suggesting that changes in redox are caused by changes in the culture conditions.
