The checkpoint protein MAD2 and the mitotic regulator CDC20 form a ternary complex with the anaphase-promoting complex to control anaphase initiation

Article Abstract:

Anaphase initiation is held up by the spindle assembly checkpoint mechanism until alignment of all the chromosomes at the metaphase plate has occurred. A ternary complex is formed by checkpoint protein MAD2 and mitotic regulator CDC20 with the anaphase-promoting complex (APC). Control of anaphase initiation is the result. The checkpoint-mediated cell cycle stopping involves hMAD2 receiving an upstream signal to cut back on activation of APC. At least three forms of mitotic APC can be seen in vivo.

Ctf7p is essential for sister chromatid cohesion and links mitotic chromosome structure to the DNA replication machinery

Article Abstract:

The sister chromatid exchange process relies on chromosome transmission fidelity (Ctf7p). Since cohesion occurs during S phase and Ctf7p activity is notable only during S phase, this suggests its role in cohesion. Ctf7p also interacts genetically with DNA metabolism mutations, which in turn affect Ctf7p chromosome loss and temperature sensitivity.
