Rhizoremediation of trichloroethylene by a recombinant, root-colonizing Pseudomonas fluorescens strain expressing toluene ortho-monooxygenase constitutively

Article Abstract:

A study was conducted regarding effective rhizoremediation of trichloroethylene (TCE) with a recombinant root-colonizing bacterium introduced into the soil by planting wheat seeds coated with the bacterium. A TCE-degrading, rhizosphere-competent bacterium was constructed from Pseudomonas fluorescens 2-79 by chromosomal insertion of the PR1(sub 23C) genes encoding constitutive expression. The study revealed that closed microcosms degraded an average of 63% of the initial TCE in 4 days compared to the 9% of the initial TCE removed by negative controls.

Combining the hok/sok, parDE, and pnd postsegregational killer loci to enhance plasmid stability

Article Abstract:

Sequencing shake-flask experiments with fully expressed plasmid-borne cloned gene were conducted to determine the stability of the plasmid pMJR1750 with hok+/sok+, parDE+, hok+/sok+ pnd+ or hok+/sok+ parDE+. The pMJR1750-aphA-pnd-hok significantly enhanced plasmid stability. However, the use of the hok+/sok+ with parDE was more effective compared to the other single-killer systems. Furthermore, the improvement in segregational stability upon the addition of a postsegregational killer locus was mediated by the killing of plasmid-free cells.
