Sgs1: a eukaryotic homolog of E. coli RecQ that interacts with topoisomerase II in vivo and is required for faithful chromosome segregation

Article Abstract:

Using a two hybrid cloning strategy, an sgs1 protein in S. cerevisiae has been identified to be structurally similar to the Escherichia coli RecQ protein. This sgs1 protein interacts with topoisomerase II in vivo and also contains helicase signature motifs similar to those of E. coli RecQ. Mutation in Sgs1 results in increase in the number of mitotic and meiotic chromosome missegregations. A model is proposed for interaction of a helicase with a topoisomerase in chromosome segregation.

Genetic and physical mapping of telomeres in the rice blast fungus, Magnaporthe grisea

Article Abstract:

Genetic mapping was applied to telomeric restriction fragments in the case of a Magnaporthe grisea linkage map. Synthetic probe and hexanucleotide repeat TTAGGG3 segregated RFLPs as consensus segregation data was used to minimize errors. Achilles' cleavage methods showed that 12 out of 14 telomeres were genetically linked to RFLP markers, indicating incomplete representation of several chromosome ends.

A complete set of marked telomeres in Saccharomyces cerevisiae for physical mapping and cloning

Article Abstract:

A URA3 containing integrating vector with telomeric TG(1-3) sequences helps mark all telomeres in Saccharomyces cerevisiae. Physical maps of chromosomes having no telomere regions are completed using these marked telomeres. Cloning and sequencing of S. cerevisiae are performed using the marked telomeres, which also help investigate telomere structure and evolution.
