Symbiotic deficiencies associated with a coxWXYZ mutant of Bradyrhizobium japonicum
Article Abstract:
The terminal oxidase complex of Bradyrhizobium japonicum was studied by creating mutations in the coxMNOP and coxWXYZ oxidases. B. japonicum is a soil bacterium which fixes N2 in the low-O2 environment of the legume root nodule. CO difference spectra from membranes of coxMNOP mutants were similar to the wild type while bacteroid membranes of coxWXYZ mutants did not have the CO-reactive cytochrome b. The coxX mutant does not have the membrane-associated O2-binding component based on data from cyanide inhibition of oxidase activity.

arfI and arfII, two genes encoding alpha-L-arabinofuranosidases in Cytophaga xylanolytica
Article Abstract:
The study described how two genes encoding alpha-L-arabinofuranosidase (ARAF) were cloned and sequenced from C. xylanolytica. The two genes, when expressed by E. coli, were able to confer ARAF activity capable of discharging both p-nitrophenol and methylumbelliferone from the respective ARAF derivatives. It was further observed that the ARAFs in this particular experiment exhibited similarities to ARAFs from B. ovatus V975, Clostridium stercorarium, Bacillus subtilis, Streptomyces lividans, and B. ovatus .
