The Caenorhabditis elegans spe-6 gene is required for major sperm protein assembly and shows second site non-complementation with an unlinked deficiency
Article Abstract:
The crawling motility of Caenorrhabditis elegans spermatozoa is dependent on its thin cytoskeletal filaments made from a cytoskeletal protein, the major sperm protein (MSP). Studies on the effects on the viability and failure of filament assembly of the spe-6 gene led to the conclusion that the gene product is required for the normal sperm development and MSP localization. A concentration-dependent interaction between spe-6 gene and the sperm-specific gene products, eDf18 and eDf19 was found necessary for MSP assembly.

Isogenic strain construction and gene mapping in Candida albicans
Article Abstract:
Researchers designed a genotypic screen that allowed determination of a heterozygous recessive mutation at the URA 3 locus to make up for the limitations in the genetic manipulation of candida albicans that are the result of its diploid genome and a sexual life cycle. The 18 bp recognition sequence of the endonuclease I-Sce I was integrated into the hisG repeats to allow the physical mapping of disrupted genes. Site-specific cleavage of the chromosome with I-SceI indicated the placement of the combined sequences.
