Transcriptional activity of the symbiotic plasmid of Rhizobium etli is affected by different environmental conditions

Article Abstract:

The specific genes involved in nodulation and nitrogen fixation in Rhizobium species are found in a single large symbiotic plasmid (pSym). Hybridization of pSym of Rhizobium etli against 32P-labelled single-stranded complementary DNAs showed that the transcriptional activity of pSym is governed by environmental stimuli. Under aerobic conditions, transcription of discrete regions of pSym are determined by carbon or nitrogen concentration. Increased gene expression was observed under conditions of poor carbon or nitrogen sources and during microaerobiosis, indicating that oxygen was a regulator of transcription. Oxygen was also suggested as a determinant of the location of transcription in pSym.

Glutamine biosynthesis and the utilization of succinate and glutamine by Rhizobium etli and Sinorhizobium meliloti

Article Abstract:

A study was undertaken to establish whether cycling of glutamine-carbon and nitrogen occurs in wild-type Rhizobium etli and Sinorizobium meliloti strains. Glutamine synthetase (GS) is a key enzyme in the glutamine cycle, and the operation of the pathway in S. meliloti mutant strains lacking GSI, and GSII, was also studied. Glutamine-carbon and nitrogen were found to be recycled by the organisms.
