Transgene integration into the same chromosome location can produce alleles that express at a predictable level, or alleles that are differentially silenced

Article Abstract:

Cre-lox mediated recombination has been use to put a gus reporter gene exactly and reproducibly into target loci in order to control the variability of transgene expression in plants. It was found that transgene integration into the same chromosome location can produce alleles that express at a predictable level. It can also produce alleles that are silenced differentially. Silencing of the introduced transgenes may be a stochastic event occurring in transformation.

KNOX homeodomain protein directly suppresses the expression of a gibberellin biosynthetic gene in the tobacco shoot apical meristem

Article Abstract:

The KNOX homeodomain protein, Nicotiana tabacum homeobox 15 (NTH15), has been found to suppress expression of a gibberellin biosynthetic gene directly in the shoot apical meristem in tobacco. An inducible system was generated using the human glucocorticoid receptor, In it steroid treatment strictly induced NTH15 function and immediately suppressed the expression of a gibberellin (GA) biosynthetic gene. A drop in bioactive GA levels also took place.

Arabidopsis cmt3 chromomethylase mutations block non-CG methylation and silencing of an endogenous gene

Article Abstract:

Eleven loss-of-function alleles were found in the CMT3 chromomethylase gene in a screen for Arabidposis mutants that change methylation and silencing of a densely methylated endogenous reporter gene. CNG methylation was lowered at repetitive centromeric sequences. The cmt3 mutants showed enhanced expression and lowered methylation of the reporter, and especially so at non-CG cytosines. Animals have mostly CG methylation and lack CMT homologs.
