Whole-cell hybridization of Methanosarcina cells with two new oligonucleotide probes

Article Abstract:

The in-situ hybridization of the methanogenic bacterium Methanosarcina has been studied by using two new oligonucleotide probes focusing on the 16S rRNA. Methanogens are the only member of domain Archaea existing in anaerobic digesters. The methane produced in biogas reactors is produced mainly from the methanogenesis of acetate and Methanosarcina cells use acetate to produce methane. These oligonucleotide probes are therefore useful tools in describing methanogenic processes.

An improved enzyme-linked immunoabsorbent assay for whole-cell determination of methanogens in samples from anaerobic reactors

Article Abstract:

A novel method for the detection of whole cells of methanogens in digester samples or anaerobic reactors was developed based on enzyme-linked immunosorbent assay (ELISA). The ELISA method was a simple adsorption procedure involving polyclonal antibodies that distinguish between methanogenic strains. Furthermore, the strategy was applicable for the quantification of whole cells of methanogens in environmental samples.

Detection of oxytetracycline production by Streptomyces rimosus in soil microcosms by combining whole-cell biosensors and flow cytometry

Article Abstract:

Bacterial biosensors and fluorescence-activated cell sorting, or flow cytometry, were successful in detecting Streptomyces rimosus oxytetracycline production in sterile soil samples. The combination leverages the biosensors' high specificity and flow cytometry's resolution power to enhance detection; it can be used to research antibiotic producer-target organism interactions in soil.
