Single crystal polarized spectroscopy of manganese superoxide dismutase and electronic structure of the active site metal complex
Article Abstract:
The optical spectra of manganese superoxide dismutase from Escherichia coli and the electronic structure of the active site metal complex were investigated using single crystal polarized spectroscopy. Manganese superoxide dismutase from Escherichia coli crystallizes from poly(ethylene glycol) solution in the orthorhombic space group C222(sub 1). Experimental results revealed a marked dichroism of the optical absorption bands that is primarily linked to a single ligand field transition near 500 nm exhibiting a polarization ratio of greater than 10:1.

Theoretical modeling of enzyme reaction chemistry: The electron transfer of the reduction mechanism in CuZn superoxide dismutase
Article Abstract:
The first step of the copper-zinc superoxide dismutase enzymatic cycle, involving the binding of a superoxide anion, and the simultaneous detachment of His63 is investigated. Results show that the protein-solvent environment fluctuations are essential to understand the reaction mechanism, which is based on the concerted rupture of the copper-histidine coordination bond and the copper-superoxide bond in the active site.

Reactivity of superoxide anion radical with a perchlorotriphenylmethyl (trityl) radical
Article Abstract:
The biochemical characterization of the reaction of superoxide with a new trityl radical, tricarboxylate derivative of perchlorotriphenylmethyl (PTM-TC) is described. The high sensitivity of detection of PTM-TC and the high rate constant of the reaction could be applied to measure superoxide in biological systems.
