Vibrational assignment of the 4-hydroxycinnamyl chromophore in photoactive yellow protein

Article Abstract:

The infrared Raman spectra is described for the dark state of the bacterial photoreceptor containing a 4-hydroxycinnamyl chromophore, photoactive yellow protein (PYP), whose chromophore is isotopically labeled with [super 13]C at the carbonyl carbon atom or at the ring carbon atoms. The observations are presented and the complete study provides base for further research of the photocycle mechanism in PYP by vibrational spectroscopy.

Energetics and role of the hydrophobic interaction during photoreaction of the BLUF domain of AppA

Article Abstract:

The characterization of the transient intermediates of the BLUF domain of transcriptional antirepressor AppA that form the basis of its photoreaction cycle was carried out by using time-resolved measurements of thermodynamic parameters. The findings indicate that a hydrophobic interaction could be the main driving force for the formation of [(AppA-BLUF).sub.2] in the photoactivated state.

Light-induced structural changes in the active site of the BLUF domain in AppA by Raman spectroscopy

Article Abstract:

Light-dependent changes in the active site of an AppA BLUF domain by Raman spectroscopy are characterized. The structural alterations are proposed to be responsible for the transmission of the light signal in the BLUF domain.
