A specific partner for abasic damage in DNA
Article Abstract:
Non-hydrogen-bonding pyrene nucleoside triphsphate (dPTP) is efficiently and specifically inserted by DNA polymerases opposite sides that do not have DNA bases. It appears that dPTP could be used in the same way as dideoxynucleotide terminators are in sequencing, operating as a specific marker for abasic sites. At least two different polymerases can insert dPTP opposite abasic sites with good efficiency, and it may therefore be feasible to analyze naturally derived DNAs for abasic damage by using cycle sequencing.
