Transmission disequilibrium test (TDT) when only one parent is available: the 1-TDT

Article Abstract:

A new transmission disequilibrium test (TDT), the 1-TDT, has been proposed for detecting linkage between a candidate marker locus and a disease locus using genotypes of affected persons and one available parent for each affected person. The test is not biased under the null hypothesis of no linkage or association and it has been validated using real and simulated data sets. Data from different types of families can be combined. The TDT in general requires genotypes of affected persons and their parents and is a method for locating mutations linked to disease genes associated with complex diseases.

A new method for estimating the risk ratio in studies using case-parental control design

Article Abstract:

Researchers announced a new method to estimate the risk ratio in studies using case-parental control design. The method was compared with Khoury's method and Flanders and Khoury's method and with Schaid and Sommer method. Researchers discovered that the new estimation method is usually smaller than that of Khoury's method or Flanders and Khoury's method, and that it is larger than Schaid and Sommer's method. When genotypic data for only one parent is available, they introduced a method to estimate the risk ratio without assuming Hardy-Weinberg equilibrium or random mating.
