Selective COX-2 inhibitors: is the water becoming muddy?
Article Abstract:
The selective cyclooxygenase (COX) inhibitor theory states that the toxicity of non-steroidal anti-inflammatory drugs in the gastrointestinal and renal systems is related to the lack of selectivity of these drugs with respect to the inhibition of COX-1 and COX-2. This hypothesis has two central tenets: the prostaglandins that are involved in pain, fever and inflammation are produced exclusively via COX-2, and that prostaglandins that play key roles in gastrointestinal functions are produced by COX-1. The accuracy of these two tenets are discussed.
