An 'anatomical cascade hypothesis' for Alzheimer's disease
Article Abstract:
Studies on individuals afflicted with Alzeimer's disease suggest that the disease process is spread by way of the neuronal pathway, beginning with the formation of neuritic plaque in the limbic system. The deposition of focal beta-amyloid precursor protein (APP), believed to be neurotoxic, causes a neuritic response that results in an upregulation of proteins responding to stress, either through heat-shock or interleukin- mediated events. Since heat-shock and interleukin-responsive elements are contained in promoters of APP-encoding genes, the APP-gene can be expected to be upregulated as a consequence of neurotoxic events.

'Traditional' pharmacotherapy may succeed Alzheimer's disease
Article Abstract:
The brain of patients with Alzheimer's disease was shown to exhibit an increase in 5-hydroxytryptamine(sub 1A) (5-HT) receptors on corticocortical pyramidal neurones. It was suggested that partial agonists acting at glutamate receptors and compounds active at the 5-HT(sub 1A) receptor should be studied to evaluate their possible therapeutic importance in the treatment of Alzheimer's disease. These traditional remedies may be valuable in salvaging functional activities lost during the slow pathological progression of Alzheimer's disease.

Mother's legacy: mitochondrial DNA mutations and Alzheimer's disease
Article Abstract:
New research findings indicate a relationship between inheritable mitochondrial DMA (mtDNA) mutations and Alzheimer's disease (AD). Mitochondria taken from the platelets of donors with AD and mtDNA deficient cells were examined. The findings suggest that mtDNA,which is inherited only through maternal lines can be the source since the mutations are more predominant in children of mothers with AD than those with fathers with AD. Evidence supporting the findings are cited.
