Synthesis and biological properties of sequence-specific DNA-alkylating pyrrole-imidazole polyamides

Article Abstract:

A series of sequence specific alkylating N-methylpyrrole (Py) and N-methylimidazole (Im) polyamide conjugates that selectively alkylate predetermined DNA sequences are designed and synthesized. Sequence-specific alkylating agents are demonstrated to possess gene-silencing activities and a promising potency against human cancer cell lines as well as against xenografts of human cancer cell lines.

Domain relationships in thiamine diphosphate-dependent enzymes

Article Abstract:

Three-dimensional structures were determined for 13 different enzymes that use thiamine diphosphate (ThDP) as a cofactor, which revealed that the enzymes fall into five families where members within a family have similar structures. A study explores the structural relationships within and between domains of ThDP-dependent enzymes.
