A role for Rac in Tiam1-induced membrane ruffling and invasion
Article Abstract:
A research study was conducted to isolate the Tiam1 gene that induces invasion and encodes proteins with a DH domain. Tiam1 is shown, in vitro, to be a GDS protein for Rho-like GTPases. The gene is also found to induce a similar phenotype as as constitutively activated (V12)Rac1 in fibroblasts, including membrane ruffling. The dominant negative (N17)Rac1 is shown to have an inhibiting effect. T-lymphoma cells that express V12Racl are found to exhibit invasiveness, which indicates that the Tiam1-Rac signalling pathway may be operating in tumor cell invasion and metastasis.

The 2.0A crystal structure of a heterotrimeric G protein
Article Abstract:
The system of the nucleotide-dependent engagement of the alpha and beta-gamma subunits of a heterotrimeric guanine-nucleotide-binding (G) protein is apparent in the structure of this protein. The refined 2.0A crystal structure of a heterotrimeric G-protein complex is analyzed. The study seeks to provide a basis for further studies that test propositions concerning the mechanism of nucleotide interaction and examines more closely how G(sub beta-gamma) promotes this process.
