A siRNA-based microbicide protects mice form lethal herpes simplex virus 2 infection

Article Abstract:

Vaginal instillation of small interfering RNAs (siRNA) targeting HSV-2 protects mice from lethal infection. siRNAs mixed with lipid are efficiently taken up by epithelial and lamina propria cells and silence gene expression in the mouse vagina and ectocervix for at least nine days and these results suggest that siRNAs are attractive candidates for the active component of a microbicide designed to prevent viral infection or transmission.

Dissecting self-renewal in stem cells with RNA interference

Article Abstract:

Genetic mechanisms controlling self-regulation in mouse embryonic stem (ES) cells are identified using an integrated functional genomics approach. Short hairpin RNA (shRNA) loss-of-function techniques are used to downregulate a set of gene products whose expression patterns suggest self-renewal regulatory functions.

Stealth RNAi directs differentiation of mouse embryonal carcinoma cells and embryonic stem cells

Article Abstract:

The power of RNAi to silence genes responsible for key developmental pathways and to direct differentiation of stem cells is demonstrated. Stealth RNAi second-generation RNAi compounds overcome the limitations of traditional small interfering RNAs by combining increased stability and specificity.
