Assignment of circadian function for the Neurospora clock gene frequency

Article Abstract:

Circadian rhythms are driven by endogenous oscillators, and synchrony with zeitgeber comes from entrainment of the oscillator. Inputs can be circadianly regulated, with outputs fed back on the oscillator. Under or overexpression of a gene product can lead to arrhythmicity. Traditional circadian entrainment protocols were used on a genetic model system, Neurospora crassa. The FRQ protein was not required for entraining an oscillator with generic circadian properties, but the circadian clock cannot synchronize to light:dark cycles without FRQ.

A mammalian gene with introns instead of exons generating stable RNA products

Article Abstract:

Seven fibrillarin-associated snoRNAs have been found to be encoded within introns of the compact mammalian U22 host gene (UHG). The unique snoRNAs, named U25-U31, show extensive complementarity to various segments of the mature rNAs. The spliced UHG RNA is poorly conserved between human and mouse, is not promising for protein coding and has a short life, showing that introns specify the functional products of UHG.

Issues are presented concerning the observation of rhythmic expression of the mPer1 clock gene in a mouse's brain. The monitoring of real-time gene expression in the transgenic mouse is discussed.
