Defective DNA single-strand break repair in spinocerebellar ataxia with axonal neuropathy-1

Article Abstract:

Tyrosyl phophodiesterase 1 (TDP1) is required for repair of chromosomal single-strand breaks in human cells arising independently of DNA replication from abortive top1 activity or oxidative stress. It is reported that TDP1 is sequestered into multi-protein single-strand break repair (SSBR) complexes by direct interaction with DNA ligase IIIalpha and that these complexes are catalytically inactive in spinocerebellar ataxia with axonal neuropathy-1 (SCAN1) cells.

Completing the map of human genetic variation

Article Abstract:

The National Human Genome Research Institute (NHGRI) has launched a community resource project that sequences large-insert clones from many individuals, systematically discovering and resolving the complex variants at the DNA sequence level. The sequence resolution of both single-nucleotide and larger-scale genomic variants will help in understanding the natural variation in human populations and the link between genetics and human health.

Specific killing of BRCA2-deficient tumours with inhibitors of poly(ADP-ribose) polymerase

Article Abstract:

A study conducted shows that poly(ADP-ribose) polymerase (PARP1) inhibitors trigger gamma-H2AX and RAD51 foci formation. It proposes that, in the absence of PARP1, spontaneous single-strand breaks collapse replication forks and trigger homologous recombination for repair.
