Article Abstract:
The study identifies the mouse Fbxw7 gene as a p53-dependent tumour suppressor gene by using a mammalian genetic screen for p53-dependent genes involved in tumorigenesis. Further, it proposes that p53-dependent loss of Fbxw7 leads to genetic instability by mechanisms that involve the activation of Aurora-A, providing a rationale for the early occurrence of these mutations in human cancers.

Researches show that the impairment of normal parental imprinting might interfere with cellular differentiation and thereby increase the probability of cancerous development. The findings also reveal that mutation of a 'cancer gene' adenomatous polyposis coli (APC) and an epigenetically imposed delay in cell differentiation might drive tumor development synergistically.
