Insights into E3 ligase activity revealed by a SUMO-RanGAP1-Ubc9-Nup358 complex

Article Abstract:

The 3.0-angstrom crystal structure of a four protein complex of Ubc9, a Nup358/RanBP2 E3 ligase domain and SUMO-1 conjugated to the carboxy-terminal domain of RanGAP1 is described. The structural insights combined with biochemical and kinetic data obtained with additional substrates, support a model in which Nup358/RanBP2 acts as an E3 by binding both SUMO and Ubc9 to position the SUMO-E2 thioester in an optimal orientation to enhance conjugation.

A down-to-earth approach

Article Abstract:

An alternative approach, which has used inhomogeneous magnetic fields, is described that has established a tunable gravity environment for crystal growth, thus recreating space on Earth. The different methods for optimizing crystal growth, as well as new synchrotron X-ray and neutron sources and microfluidics and robotics have helped in the analysis of biological crystal structures.

Structure of the ESCRT-II endosomal trafficking complex

Article Abstract:

The crystal structure of the core of the yeast ESCRT-II complex, which contains one molecule of the Vps protein Vps22, the carboxy-terminal domain of Vps36 and two molecules of Vps25 and has the shape of capital letter 'Y' is presented. The 3.6 angstorm resolution structure of the quarternary ESCRT-II complex containing Vps22, Vp36 and two Vps25 molecules are reported.
