Laboratory evolution of peroxide-mediated cytochrome P450 hydroxylation

Article Abstract:

The desire to create practical single-enzyme hydroxylation catalysts has prompted efforts to create cytochrome P450s that efficiently use hydrogen peroxide instead of dioxygen and NADH. This work has involved the directed evolution of the P450 from Pseudomonas putida to form mutants that hydroxylate naphthalene where no cofactors are present through the 'peroxide shunt' pathway. Activity is more than 20-fold higher than the native enzyme. Further evolution of P450s would make it possible to investigate the limits of P450-catalyzed aromatic hydroxylation through the peroxide shunt pathway.

A synthetic multicellular system for programmed pattern formation

Article Abstract:

A synthetic multicellular system in which genetically engineered 'receiver' cells are programmed to form ring-like patterns of differentiation based on chemical gradients of an acyl-hormoserine lactone (AHL) signal that is synthesized by 'sender' cells is demonstrated. In receiver cells, 'band-detect' gene networks respond to user-defined ranges of AHL concentrations.

Programmed population control by cell-cell communication and regulated killing

Article Abstract:

The population-control circuit laid the foundation for using cell-cell communication to programme interactions among bacterial communities. This allows the concept of communication-regulated growth and death to be extended to engineering synthetic ecosystems.
