Inhibition of nociceptors by TRPV1-mediated entry of impermeant sodium channel blockers

Article Abstract:

The charged sodium-channel blockers can be targeted into nociceptors by applying TRPV1 agonists in order to produce a pain-specific local anesthesia. Injection of QX-314 together with capsaicin into rat has produced a long-lasting increase in mechanical and thermal nociceptive thresholds, but has not shown any motor or tactile defects.

Voltage-sensor activation with a tarantula toxin as cargo

Article Abstract:

The kinetics and state dependence of the toxin-channel interaction and the physical location of the toxin in the membrane are examined. The formation of a strong and stable complex of voltage sensors with hanatoxin and the reduction of toxic-affinity is observed due to voltage-sensor activation.
