Research describing the processes of DNA replication forks is presented. In particular the effects of the DNA-alkylating agent methyl methanesulphonate are investigated.

A Mec-1 and Rad53-dependent checkpoint controls late-firing origins of DNA replication
Article Abstract:
It is shown that hydroxyurea in budding yeast, not only blocks the progression of replication forks from early-firing origins, but also inhibits the firing of late origins. These are maintained in an initiation-competent prereplicative state for long periods. The active process of blocking to late origin firing is yeast defective with mutations in rad53 and mec1 checkpoint genes.
