Requirement of phosphatidylinositol 4,5-biphosphate for alpha-actinin function

Article Abstract:

Alpha-actinin, a protein from striated muscle, is an endogenous phosphatidylinositol 4,5-biphosphate (PtdInsP2). Moreover, PtdInsP2 controls the F-actin-gelating activity of alpha-actinin by interacting with it. The ability of PtdInsP2 to boost the F-actin-gelating activity of alpha-actinin in smooth muscles to levels normally observed only in striated muscles indicates that PtdInsP2 is requisite for alpha-actinin to maximize its gelating activity.

Induction of filopodium formation by a WASP-related actin-depolymerizing protein N-WASP

Article Abstract:

Cdc42 is a GTPase in the Rho family and several proteins bind Cdc42 directly, although none can account for filopodium formation induced by Cdc42. An experiment shows that Cdc42 must bind a WASP-related protein, N-WASP before it can induce filopodium formation. Addition of active Cdc42 was shown to significantly stimulate N-WASP actin-depolymerizing activity, in a cell-free system.
