Therapeutic silencing of an endogenous gene by systemic administration of modified siRNAs

Article Abstract:

Chemically modified short interfering RNAs (siRNAs) silence endogenous gene encoding apolipoprotein B (apoB) after intravenous injection in mice. Administration of chemically modified siRNAs result in silencing of the apoB messenger RNA in liver and jejunum, decrease plasma levels of apoB protein, and also reduce total cholesterol.

A cholesterol connection in RNAi

Soutschek et al. describes a simple but effective method for the intravenous delivery of nucleic acids called short interfering RNAs that target a therapeutically important messenger RNA molecule. The point at which the mRNA is severed is identified.
