Automated purification and suspension array detection of 16S rRNA from soil and sediment extracts by using tunable surface microparticles

Article Abstract:

A study is conducted to simply the fluidic requirements for integrated biodetection system with the help of a tunable surface microparticles both as rRNA affinity purification resin in a renewable microcolumn sample preparation system and as the sensor surface in a flow cytometer detector. The implications of unexpected target cross-hybrization and fluorescence quenching are discussed relative to the design and implementation of an integrated microbial monitoring system.

Miniprimer PCR, a new lens for viewing the microbial world

Article Abstract:

A new polymerase, S-Tbr, and short primers are used to show the utility of a 'miniprimer' PCR method. Deeply divergent sequences are discovered with high frequency and included representatives that define two new division-level taxa, which has shown that miniprimer PCR method might disclose new dimensions of microbial diversity.

Genomic DNA amplification from a single bacterium

Article Abstract:

Genomic DNA was amplified about 5 billion-fold from single, flow-sorted bacterial cells by the multiple displacement amplification (MDA) reaction, using phi 29 DNA polymerase. A 662-bp segment of the 16S rRNA gene could be accurately sequenced from the amplified DNA.
