Characterization of 16S rRNA genes from oil field microbial communities indicates the presence of a variety of sulfate-reducing, fermentative, and sulfide-oxidizing bacteria

Article Abstract:

Cloning and nucleotide sequencing experiments are performed on PCR-amplified 16S rRNA genes to characterize oil field microbial populations. Novel microorganisms are isolated and their chromosomal DNAs are used in analyzing free-floating and attached microbial communities in the oil field environment. Results indicate the presence of a variety of gram-negative, sulfate-reducing bacteria and a more limited number of anaerobic, fermentative and acetogenic bacteria.

Effect of nitrate injection on the microbial community in an oil field as monitored by reverse sample genome probing

Article Abstract:

A modified form of reverse sample genome probe (RSGP) method was developed for the isolation of heterotrophic bacteria from oil fields. The RSGP method can be utilized to monitor the response of microbial communities to environmental changes such as high-potential electron acceptor nitrate availability. The quantitation of hybridization signals was also enhanced by utilizing phosphorimaging plate technology. Furthermore, RSGP analysis indicated the inability of the heterotropic bacteria from the master filter to become dominant community components after the injection of nitrate.
