Development and application of real-time PCR assays for quantification of erm genes conferring resistance to macrolides-lincosamides-streptogramin B in livestock manure and manure management systems

Article Abstract:

The development, validation and use of six real-time PCR assays for quantifying six classes of erm genes that encode the major mechanism of resistance of macrolides-lincosamides-streptogramin B (ML[S.sub.B]) is reported. It is suggested that the methods of manure storage and treatment has a substantial impact on the persistence and decline of (ML[S.sub.B]) resistance originating from food animals that affects the dissemination of resistance genes into the environment.

Comparisons of different hypervariable regions of rrs genes for use in fingerprinting of microbial communities by PCR-denaturing gradient gel electrophoresis

Article Abstract:

The PCR-denaturing gradient gel electrophoresis (DGGE) profiles arising from a common set of community DNA samples, using the previously defined primer sets, are compared. The result shows that DGGE analysis of gastrointestinal mocrobiomes is best accomplished by the amplification of either the hypervariable (V) 3 or V1 region of rrs genes.

Evaluations of different hypervariable regions of archaeal 16S rRNA genes in profiling of methanogens by Archaea-specific PCR and denaturing gradient gel electrophoresis

Article Abstract:

Different hypervariable (V) regions of the archaeal 16S rRNA gene (rrs) are compared to establish a preferred V region for using in Archaea-specific PCR-denaturing gradient gel electrophoresis (DGGE). The analysis has shown that different methanogens are detected when different V regions are targeted by PCR-DGGE.
