Electron and fluorescence microscopy of extracellular glucan and aryl-alcohol oxidase during wheat-straw degradation by Pleurotus eryngii

Article Abstract:

Research was performed to analyze aryl-alcohol oxidase (AAO) distribution during differential degradation by Pleurotus eryngii. Immunofluorescence microscopy and transmission electron microscopy were used to observe the conditions governing the secretion of extracellular polysaccharide and AAO as well as straw degradation during solid-state fermentation. Results revealed that P. eryngii produces a hyphal sheath consisting of a thin glucan layer and that AAO penetrates the plant cell wall during P. eryngii degradation of wheat straw.

Regulation of peroxidase transcript levels in liquid cultures of the ligninolytic fungus Pleurotus eryngii

Article Abstract:

Peroxidase transcript levels in liquid cultures of the ligninolytic fungus Pleurotus eryngii and their regulation are discussed. Northern blotting was used for studying them. These peroxidases seem different from P. chyrsosporium LiP and MnP in molecular structure and catalytic properties with expression apparently regulated differently. Regulation is by N source, Mn(super.2+), and oxidative stress of the transcript levels of the unique peroxidase produced where liquid cultures of P. eryngii with peptones are used.

Transformation of industrial dyes by manganese peroxidases from Bjerkandera adusta and Pleurotus eryngii in a manganese-independent reaction

Article Abstract:

Research was conducted to examine the transformation of industrial dyes from Bjerkandera adusta and other white rot fungi by ligninolytic peroxidases. A comparison was made with the transformation of dyes by MnP isoenzymes from Phanerochaete eryngii and P. chrysosporiium. Results indicate that some differently substituted sulfonated azo dyes were better oxidized in the presence of Mn2+ at pH 5 while others were better oxidized in the absence of Mn2+ at pH 3.
