Extracellular production of neoculin, a sweet-tasting heterodimeric protein with taste-modifying activity, by Aspergillus oryzae

Article Abstract:

The extracellular production of recombinant neoculin (rNCL) with sensory activities by Aspergillus oryzae is reported and an efficient strategy is designed to express the two heterodimeric subunits with different productivities and increased rNCL production level by constitutive induction of the unfolded protein response (UPR). The analysis has shown that the A. oryzae production system is useful for the production of heterooligomeric proteins as well as for probing the molecular biology of this unique taste-modifying protein.

Isolation and characterization of Aspergillus oryzae vacuolar protein sorting mutants

Article Abstract:

The vacuolar protein sorting (vps) system in the filamentous fungus Aspergillus oryzae which has unique cell polarity and the ability to secrete large amounts of proteins were evaluated by using mutants that missort vacuolar proteins into the medium. It is suggested that vacuolar carboxypeptidase Y and green fluorescent protein (CPY-EGFP) are transported by a novel pathway possibly involving the Spitzenkorper.

Molecular analysis of an inactive aflatoxin biosynthesis gene cluster in Aspergillus oryzae RIB strains

Article Abstract:

A study was conducted on the molecular analysis of the structure of an aflatoxin biosynthesis gene homolog cluster Aspergillus oryzea RIB 40, to help assess the potential for aflatoxin production by A. oryzae. The results showed further classification of the A. oryzae RIB strains into three groups, based on the PCR results.
