Heterologous production of functional forms of Rhizopus oryzae lipase in Escherichia coli

Article Abstract:

The expression of the Rhizopus oryzae lipase (ROL) gene in Escherichia coli is presented for the time to yield a correctly folded product, present only in the cytoplasm fraction. The results have shown that the production of active ROL and its precursor ProROL in soluble form is achieved when Escherichia coli Origami (DE3) and pET-11d are used as expression systems.

Enhancement of the stability of a prolipase from Rhizopus oryzae toward aldehydes by saturation mutagenesis

Article Abstract:

Prolipase from Rhizopus oryzae (proROL) is engineered in order to enhance its stability toward lipid oxidation products like aldehydes with the aim of boosting its performance in oleochemical industries. A microtiterplate (MTP)-based assay is designed for determining the inactivation kinetics and also identify more stable, but highly active variants.
