IS1236, a newly discovered member of the IS3 family, exhibits varied patterns of insertion into the Acinetobacter calcoaceticus chromosome

Article Abstract:

Analysis of the IS1236 nucleotide sequence in Acinetobacter calcoacetius, shows biochemical characteristics that identify it to be a member of the IS3 family with the greatest sequence similarity over ORFB. There is a significant divergence in the ORFA amino acid sequence. IS1236 insertion, mutations are evenly distributed in pobR and pcaH. The sites of insertion in pobR are flanked by a 3 bp DNA sequence repetition. However, in pcaH duplication patterns are different and indicate that insertion patterns are influenced by IS1236 DNA target.

The recA gene from Clostridium perfringens is induced by methyl methanesulphonate and contains an upstream Cheo box

Article Abstract:

The biochemical properties of the recA gene from Clostridium perfringens were analyzed to determine the genetic mechanisms involved in microbial homologous recombination and DNA repair. Analysis of Clostridium perfringens recA gene indicated the role of DNA damage in activating the RecA protein due to exposure of the cells to methyl methanesulphonate. Furthermore, induction of recA gene due to DNA damage in Clostridium perfringens was also mediated by the presence of an SOS-like Cheo box operator in the upstream region.

The expression of the Acinetobacter calcoaceticus recA gene increases in response to DNA damage independently of RecA and of development of competence for natural transformation

Article Abstract:

The transcriptional regulation of the Acinetobacter calcoaceticus recA gene was analyzed by employing the lacZ operon fusion method. There was a low inductive capacity for compounds that damage DNA and/or prevent DNA replication. The presence of the recA promoter region on a multicopy plasmid had the same effect on recA transcription as the presence of DNA-damaging agents. Therefore, recA expression in A. calcoaceticus may not be part of a regulon responsible for competence for natural transformation.
