Loss of transcriptional activity of a transgene is accompanied by DNA methylation and histone deacetylation and is prevented by insulators

Article Abstract:

A DNA sequence containing the constitutive DNase I hypersensitive site at the 5' end of the chicken beta-globin locus acts as an insulator. The chicken insulator can protect a stably integrated gene, for which there is otherwise great variability in expression, from chromatin-mediated repression in cell culture. It protects against position effects in Drosophila. A model has been proposed for the chicken insulator's ability to protect against extinction in the transformed cell lines and to function as a chromatin boundary for the chicken beta-globin locus in normal erythroid cells. Transcriptional activity loss in the transgene takes place with DNA methylation and histone deacetylation. It is prevented by insulators.

Seven-up, the Drosophila homolog of the COUP-TF orphan receptors, controls cell proliferation in the insect kidney

Article Abstract:

Morphogenesis of the Malpighian tubules in insects is controlled in Drosophila by the 'tip cell,' which is one large cell. Seven-up is an important component induced by mitogenic epidermal growth factor (EGF) receptor signaling from the tip cell. Seven-up can then regulate transcription of regulators of the cell cycle. The tip cell has been thought to send a mitogenic signal that brings on division in close-by cells. Seven-up is a homolog of the human transcription factor COUP-TF.

Clockwork orange is a transcriptional repressor and a new Drosophila circadian pacemaker component

Article Abstract:

Clockwork orange (cwo) and the master clock gene Clock from Drosophila which promotes the transcription of several key target genes were studied. The results indicate that cwo acts preferentially in the late night to help terminate CLK-CYC mediated transcription of direct target genes including cwo itself.
