Molecular microbial diversity of an agricultural soil in Wisconsin

Article Abstract:

Microbial diversity of an agricultural soil in southern Wisconsin was determined by sequence analysis of clones of PCR-amplified DNA extracted from the soil. A total of 98.4% of the 124 clones sequenced came from the domain Bacteria. Two of the clones were of nuclear origin, one fungal and the other a plant sequence. The domain Archaea was not represented. Sequences from the kingdoms Proteobacteria, the Cytophaga-Flexibacter-Bacteroides group and the low-G+C-content gram-positive group were most prominent among the domain Bacteria. No sequences from the kingdoms Thermotogales, Fusobacteria and Spirochaetes were present.

Bacterial rRNA genes associated with soil suppressiveness against the plant-parasitic nematode Heterodera sahachtii

Article Abstract:

Research focuses on the identification of bacteria involved in the Heterodera schachtii suppressiveness by correlating population level with the suppressiveness. Polymerase chain reaction analyses of the bacterial population directed to H. schachtii cysts in soil identifies one bacterial rDNA group with sequence identity to rDNA from Rhizobium species, suggesting its involvement with the H. schachtii suppressiveness.

PCR primers that amplify fungal rRNA genes from environmental samples

Article Abstract:

Research examines deployment of recombinat DNA techniques in the analysis of environmental samples containing biological material. Construction of polymerase chain reaction primers for amplifying rRNA genes of fungi is described. Data show that using these tools, soil samples can be analyzed for their fungal composition not detected by the conventional culture techniques.
