Multiple cis-acting sequences contribute to evolved regulatory variation forDrosophila Adh genes

Article Abstract:

The alcohol dehydrogenase (Adh) genes of Drosophila affinidisjuncta and D. hawaiiensis are expressed in a species- specific manner. To study the basis of this differential regulation, cis-actingsequences contributing to the regulatory differences were mapped. Three gene segments in the promoter region were found to have large and specific effects, while other portions of the genes have smaller, quantitative effects. Some chimeric genes displayed both tissue-specific and systemic changes in RNA levels. These results suggest that the cis-acting sequences are involved in both Adh-specific expression and in tissue-specific regulatory differences.

Delineation of cis-acting sequences required for expression of Drosophila mojavensis Adh-1

Article Abstract:

The regulation of the alcohol dehydrogenase (Adh) gene of Drosophila mojavesis was elucidated with respect to possible upstream regulatory sequences. Deletion studies showed that correct larval expression of Adh-1 is determined by a sequence 70 bp upstream of the transcriptional start site. Further localizationof this cis-acting element placed it downstream of -100 but upstream of -80. This region was found to contain a sequence neccessary for Adh-1 expression in the ovary.

Rearrangement of upstream regulatory element leads to ectopic expression of the Drosophila mulleri Adh-2 gene

Article Abstract:

A study was conducted to examine the ectopic expression of the Adh-2 gene of Drosophila mulleri. The gene is expressed in the larval fat and adult fat bodies and hindgut. Maximum transcription requires a 1500-bp element 2-3 kb upstream of the Adh-2 promoter. Earlier findings indicate that deletion sequences between this promoter and the upstream element expresses Adh-2 genes in the middle midgut. Results show that sequence elements control ectopic expression patterns.
