Multiple female reproductive failures in cyclooxygenase 2-deficient mice

Article Abstract:

Experiment was conducted to show that the targeted disruption of cyclooxygenase (COX)-2, but not COX-1 in mice generates multiple failures in female reproductive processes that include ovulation, fertilization, implantation and decidualization. COX is the rate-limiting enzyme in the synthesis of prostaglandins and exists in two isoforms, COX-1 and COX-2. Multiple approaches were used in the study. Results suggest that defects result from the deficiency of pituitary gonadotropins or ovarian steroid hormones, or reduced responsiveness of target organs to respective hormones.

Intestinal tumorigenesis in compound mutant mice of both Dpc4 (Smad4) and Apc genes

Article Abstract:

Research was conducted to study the role of the DPC4 mutation in carcinogenesis and establish germ-line transmitted mutant mice. Tissue specimens were harvested according to standard protocols while pDpc 4 was linearized at a single site. Results showed that some of the cis-compound heterozygotes developed ampullary carcinomas in the duodenal papilla and that more than one half of the heterozygotes had epidermoid cysts of the skin.

Suppression of intestinal polyposis in APCdelta716 knockout mice by inhibition of cyclooxygenase 2 (COX-2)

Article Abstract:

The influence of the cycloxygenase 2 (COX-2) isozyme in colorectal tumorigenesis is investigated. The effects of COX-2 gene knockouts and a unique COX-2 inhibitor on Apcdelta16 knockout mice are analyzed. Data indicate that COX-2 has a significant role in tumorigenesis and that COX-2-selective inhibitors can act as unique therapeutic agents for colorectal polyposis and cancer.
