Novel paired starter culture system for sauerkraut, consisting of a nisin-resistant Leuconostoc mesenteroides strain and a nisin-producing Lactococcus lactis strain

Article Abstract:

A paired starter culture system was evaluated in order to improve the fermentative process of producing sauerkraut. A nisin-resistant Leuconostoc mesenteroides was able to grow well in the presence and absence of a nisin-producing Lactococcus lactis subsp lactis. Similarly, L. lactis subsp lactis was able to grow in the presence and absence of Leuconostoc mesenteroides. The optimum ratio of L. lactis subsp lactis to Leuconostoc mesenteroides was 1,000 is to 1. The mixed culture was able to retard the growth of a nisin-sensitive strain of Lactobacillus plantarum.

Characterization of two nisin-producing Lactococcus lactis subsp. lactis strains isolated from a commercial sauerkraut fermentation

Article Abstract:

Screening of isolates collected from sauerkraut fermentation yielded two Lactococcus lactis subsp. lactis strains, NCK400 and LJH80, which were able to produce nisin. LJH80 was morphologically unstable and gave rise to to nisin-producing derivatives which could be distinguished by their plasmid and genomic DNA profiles. NCK400 did not harbor plasmids, indicating that the nisin structural gene was chromosomally integrated. Hybridization patterns using the structural gene for nisin as probe confirmed that the isolates obtained in this study were unique.

IS946-mediated integration of heterologous DNA into the genome of Lactococcus lactis subsp. lactis

Article Abstract:

The use of the integration vector from the lactococcal insertion sequence IS946 for the chromosomal insertion of heterologous DNA into the Lactococcus lactis subspecie lactis was found to be avery efficient method for recovering integration events. The vector used was constructed from the lactococcal insertion sequence IS946. Its construction provides an additional tool for the genetic analysis and manipulation of lactococci.
