Oligomeric complexes link Rab5 effectors with NSF and drive membrane fusion via interactions between EEA1 and syntaxin 13

Article Abstract:

A study was conducted to determine the molecular links between the Rab mechanism controlling early endosome tetherng and SNARE-mediated fusion. Results showed that the Rab5 effectors EEA1 and Rabaptin-5/Rabex-5 are present on the membrane of high molecular weight oligomers that also contain NSF. The oligomeric assembly was regulated by the ATPase activity of NSF. A direct interaction between EEA1 and syntaxin 13, the t-SNARE required for endosome fusion, was also observed, suggesting that this interaction is required to drive endosome fusion.

Modulation of receptor recycling and degradation by the endosomal kinesin KIF16B

Article Abstract:

The kinsein-3, KIF16B, transports early endosomes to the plus end of microtubules in a process regulated by the small GTPase Rab5 and its effector, the phosphatidylinositol-3-OH kinase hVPS34. The results reveal that KIF16B, by regulating the plus end motility of early endosomes, modulates the intracellular localization of early endosomes and the balance between receptor recycling and degradation.

APPL proteins link Rab5 to nuclear signal transduction via an endosomal compartment.

Article Abstract:

A pathway directly linking the small GTPase Rab5, a key regulator of endocytosis, to signal transduction and mitogenesis is identified. Findings reveal an endosomal compartment bearing Rab5 and APPL proteins as an intermediate in signaling between the plasma membrane and the nucleus.
