Organization of the European Bacillus subtilis genome sequencing project

Article Abstract:

The European Commission supports genome sequencing of Bacillus subtilis under the biotechnology program. The project involves development of the physical map, generation of suitable DNA libraries and the systematic sequencing of the B. Subtilis genome. Cloning in plasmid, lambda and YAC vectors, in vitro amplification with PCR, ordering of lambda clones using YAC recombinant vectors and the use of integrative plasmids and marker rescue in genome walking are some of the methods used to generate ordered sublibraries. lys A, gly C, pks X, tyr Z, orf X17, orf X18, spoV AF and ipa-16d are some of the putative genes determined during the initial phase of the project.

SubtiList: a relational database for the Bacillus subtilis genome

Article Abstract:

SubtiList, a relational database for the management and analysis of information related to the molecular genetics of Bacillus subtilis, permits the retrieval of non-redundant DNA sequences of the bacterium. A logical structure provides suitable links between the different objects, while a series of steps enable data updating and management, and a user-friendly interface permits easy access. A DNA searching program and a program for fast similarity searching of protein data banks are a few of the sequence analysis tools available.

The European Bacillus subtilis genome sequencing project: current status and accessibility of the data from a new World Wide Web site

Article Abstract:

The genome sequencing projects for Bacillus subtilis by European and Japanese research groups are vital for the molecular research and analysis of genome evolution and bacterial cell physiology. The European deoxyribonucleotide sequence database, SubtiList, has been constantly updated and it already contains 3311 kb of non-redundant ribonucleic acid. The SubtiList database also contains 79% of the 4188 kb chromosome which were also derived from Japanese sequencing networks.
