Overexpression of bacterial hemoglobin causes incorporation of pre-beta-lactamase into cytoplasmic inclusion bodies

Article Abstract:

The resulting fusion of pre-beta-lactamase into cytoplasmic inclusion bodies caused by the overexpression of Vitreoscilla hemoglobin is discussed. Pre-beta-lactamase was aggregated from Escherichia coli and its activity was measured with an ampicillin substrate. Polyacrylamide gel electrophoresis, immunoblotting, plasmid preparation and transmission electron microscopy were performed in the experiment. Results indicate that the extent of in vivo aggregation of a recombinant protein within a bacterial cell is also affected by the coexpression of another recombinant protein.

Entry of Escherichia coli into stationary phase is indicated by endogenous and exogenous accumulation of nucleobases

Article Abstract:

Escherichia coli cells entering the stationary growth phase excrete uracil and xanthine into the culture medium and increase the amount of intracellular free nucleobases. Hypoxanthine is the main intracellular nucleobase while xanthine is present in small amounts. The endogenous and exogenous accumulation of the nucleobases is due to the degradation of rRNA. The bacterial cells normally contain nucleotides rather than nucleobases.

Physiology and metabolic fluxes of wild-type and riboflavin-producing Bacillus subtilis

Article Abstract:

The energy metabolism of growth and product synthesis in wild-type and riboflavin-producing Bacillus subtilis from chemostat cultures was investigated by analytical techniques, amino acid analysis and measurement of flux rate. Both strains exhibited identical growth yields. There was a significant relationship between riboflavin production rate and dilution rate, suggesting that product formation is an indicator of growth.
