Promoter cleavage: A Topollbeta and PARP-1 collaboration

Article Abstract:

The estrogen-dependent restructuring and transcription of the pS2 promoter requires the generation of a DNA double-strand break by a novel protein complex containing two enzymes, TopoIIbeta and PARP-1. The enzymes topoisomerase IIbeta (Topollbeta) and poly(ADP-ribose) polymerase (PARP-1) have collaborated in a way that alters the molecular composition and structure of the pS2 promoter during an estradiol (E2)-dependent transcriptional response.

NAD+- dependent modulation of chromatin structure and transcription by nucleosome binding properties of PARP-1

Article Abstract:

The nucleosome binding properties of PARP, which is the most abundantly expressed member of the protein family that catalyze the transfer of ADP-ribose units from NAD+ to specific target proteins, are discussed. The auto modification activity of PARP-1 is stimulated by nucleosomes, which release PARP-1 from chromatin, and the NAD+- dependent activities of PARP-1 are reversed by PARG and are inhibited by ATP in vivo.

PARP goes transcription

Article Abstract:

This review examines the role of poly(ADP-ribose)polymerase in the modulation of both chromatin structure and transcription in vivo. Authors discuss poly(ADP-ribose)polymerase-1 structure, funtion, and activity, regulaion of its catalytic activity, chromatin structure and transcription, nucleosomes, genetic enhancers and promoters, and gene expression.
