'Puckered' encodes a phosphatase that mediates a feedback loop regulating JNK activity during dorsal closure in Drosophila

Article Abstract:

A subset of phosphatases, dually specific, mediate a MAP kinase (MAPK) phosphatase activity. A member of the VH-1 family coded by the puckered gene of Drosophila has been described. Mutations in the gene bring on defects that lead to imperfect dorsal closure. The mutations are related to those associated with mutation in basket, the Drosophila JNK homolog. The puckered mutations lead to the hyperactivation of DJNK. Expression of puckered comes from activity of the JNK pathway. In dorsal closure JNK signals activate an effector coded by decapentaplegic and an element of negative feedback regulation coded by puckered.

The Drosophila gene brinker reveals a novel mechanism of Dpp target gene regulation

Article Abstract:

The decapentaplegic (dpp), a Drosophila member of the TGFbeta family of secreted molecules, acts as a long-range morphogen in patterning of the embryo and the adult appendages. Dpp signals through the SMAD proteins Mad and Medea. However, it was found that in the absence of brinker (brk), Mad is not needed for the activation of Dpp target genes that depend on low levels of Dpp The brk gene encodes a novel protein the properties of a transcriptional repressor.
