RNA export

Article Abstract:

Export of RNA from the nucleus involves movement of the ribonucleoprotein (RNP) to the nuclear pore complex (NPC), followed by RNP translocation through the NPC. This transport is mediated by RNA-binding proteins, nucleoporins (NPC proteins) and RanGTPase. The RNA-binding proteins include heterogeneous RNA (hnRNA) nuclear proteins, some of which move between the nucleus and cytoplasm. Some viral proteins, such as the HIV protein Rev, have been found to play a regulatory role in RNA export. Genes that encode export factors are also discussed, as is the RanGTPase cycle.

The human U1 snRNP-specific U1A protein inhibits polyadenylation of its own pre-mRNA

Article Abstract:

The function of the human U1 small nuclear ribonucleoprotein (snRNP)-specific U1A protein during the processing of pre-mRNA was characterized. Human mRNAs encoding the U1 snRNP-specific protein contain a 47-nucleotide conserved region in its 3' untranslated region (UTR). In vitro and in vivo experiments showed that U1A protein can bind to the 3' UTR of human U1A mRNA, thus inhibiting the polyadenylation of its own pre-mRNA. This suggests a mechanism for regulating the production of U1A mRNA via pre-mRNA binding and polyadenylation.

The HIV-1 Rev activation domain is a nuclear export signal that accesses an export pathway used by specific cellular RNAs

Article Abstract:

The HIV-1 Rev activation domain is a nuclear export signal that redirects Rev response element (RRE)-containing viral RNAs to a non-mRNA export pathway. HIV-1 Rev protein directs nuclear export of pre-mRNAs and mRNAs containing its binding site, RRE. The export of mRNA, tRNA, or rRNA is unaffected by the bovine serum albumin containing Rev activation domain (BSA-R).
