Reinforcement of silencing at transposons and highly repeated sequences requires the concerted action of two distinct RNA polymerases IV in Arabidopsis

Article Abstract:

Genetic and biomedical studies have revealed the existence in plants of fourth RNA polymerase, RNAPIV, which mediates siRNA accumulation and DNA methylation-dependent silencing of endogenous repeated sequences. Study is conducted to show that Arabidopsis expresses two evolutionarily related forms of RNAPIV which are RNAPIVa and RNAPIVb and selective disruption of either forms indicates that high levels of DNA methylation and silencing depends on siRNA accumulated through pathway involving the concerted action of both RNAPIV forms.

An ARGONAUTE4-containing nuclear processing center colocalized with Cajal bodies in Arabidopsis thaliana

Article Abstract:

Study is presented to show that ARGONAUTE4 (AGO4) localizes to nucleolus-associated bodies along with the RNA polymerase IV (Pol IV) subunit NRPD 1b, which are required for DNA methylation guided by 24 nucleotide small interfering RNAs (siRNAs) in Arabidopsis thaliana. The results have indicated a function for Cajal bodies as a center for the assembly of an AGO4/NRPD1b/siRNA complex, facilitating its function in RNA-directed gene silencing at target loci.

Reiterated WG/GW motifs form functionally and evolutionarily conserved ARGONAUTE-binding platforms in RNAi-related components

Article Abstract:

A study was undertaken to show that the extended C-terminal domain of NRPD1b-type proteins exhibited conserved Argonaute-binding capacity through a WG/GW-rich area. Results revealed that WG/GW motifs formed functionally conserved Argonaute-binding platforms in RNAi-related components.
