Sec6/8 complex is recruited to cell-cell contacts and specifies transport vesicle delivery to the basal-lateral membrane in epithelial cells

Article Abstract:

A rapid recruitment of Sec6/8 complex from the cytosol to cell-cell contacts occurs when calcium-dependent adhesion in the epithelial cells is implemented. Such process results to the development of cell polarity which restricts the distribution of Sec6/8 complex to the apical-junctional complex of the lateral membrane of epithelial cells. Thus, biogenesis of epithelial cell surface polarity is influenced by the lateral recruitment of the Sec6/8 complex which resulted from cell-cell adhesion.

RalB GTPase-mediated activation of the IkB family kinase TBK1 couples innate immune signaling to tumor cell survival

Article Abstract:

The relative contribution of RalGTPase effector proteins to Ral function in normal and tumorigenic human epithelial cells were investigated in order to define the regulatory pathway directly engaged by RalB to support tumor cell survival. The study observations define the mechanistic contribution of RalGTPases to cancer cell survival and reveal the RalB/Sec5 effector complex as a component of TBK1-dependent innate immune signaling.

A new family of effectors that share WxxxE sequence motif and directly stimulate host signaling pathways by mimicking activated Ras-like cellular GTPases is described. Many bacterial pathogens use a specialized type III secretion system to deliver virulence effector proteins into host mammalian cells.
